Antithrombin III Inhibits Mesangial Cell

چکیده
Thrombin stimulates and heparin and heparan sulfate inhibit mesangial cell proliferation. In addition, heparin has been shown to inhibit thrombin-stimulated smooth muscle cell proliferation. The anticoagulant action of heparin is mediated by antithrombin Ill. This study investigated whether heparin’s antiproliferative action is also mediated by antithrombin Ill. To this end, the effect of antithrombin Ill on thrombinstimulated mesangial cell growth was examined. As expected, thrombin stimulated DNA synthesis and cell growth in cultured human mesangial cells. The effect of thrombin on DNA synthesis and mesangial cell proliferation was inhibited by standard heparin and antithrombin Ill, separately or together. The magnitude of the inhibitory action of antithrombin Ill was equal to that of equimolar concentrations of heparin and that observed with the combination of the two. Experiments carried out in serum (hence antithrombin 111)-free medium revealed that heparin’s inhibitory effects are independent of antifhrombin Ill. It was concluded that antithrombin Ill, an endogenous inhibitor of thrombin’s coagulant activity, is an equally effective inhibitor of thrombin’s mitogenic action.

Antithrombin III inhibits thrombin-induced proliferation in human arterial smooth muscle cells.
Thrombin has attracted increasing attention as a possible mitogen for vascular smooth muscle cells in lesion development both after vascular injury and in atherogenesis. In this study, the ability of antithrombin III to inhibit alpha-thrombin-induced DNA synthesis and cell proliferation in human arterial smooth muscle cells was analyzed. We demonstrate a concentration-dependent initiation of DN...
متن کاملQuantitative Cell Numbers and Density of Mesangial Volume in a Rat Model with Induced Hyperglycemic and Treated with Mononuclear Derived CD133 Positive Cells
Background: There is evidence that mesangial cell structural changes contribute to the pathogenesis of diabetic nephropathy. To gain better insight into the mechanisms responsible for this issue, present study focused on effect of cord blood mononuclear cells (MNCs) derived CD133 positive cells on mesangial cell structure and function. Materials and Methods: The animals were randomly divided in...
متن کاملEffects of argatroban as an anticoagulant for haemodialysis in patients with antithrombin III deficiency.
BACKGROUND In congenital or acquired antithrombin III-deficient patients undergoing haemodialysis, coagulation or residual blood in the blood circuit and dialyser is commonly observed under anticoagulation with heparin. Argatroban, a synthetic thrombin antagonist, directly inhibits thrombin activity in a manner that is different from that of heparin, thereby displaying an anticoagulating effect...
متن کاملHEMOSTASIS, THROMBOSIS, AND VASCULAR BIOLOGY Autoantibody against prothrombin aberrantly alters the proenzyme to facilitate formation of a complex with its physiological inhibitor antithrombin III without thrombin conversion
Acquired coagulation factor inhibitors include pathologic immunoglobulins that specifically bind to coagulation factors and either neutralize their procoagulant activity, accelerate their clearance from the circulation, or have proteolytic activity to degrade them into inactive polypeptides. Here, an autoantibody against prothrombin is described in a patient with serious hemorrhagic diatheses. ...
